忧遁草(Clinacanthus nutans)为东南亚地区的民俗药材,台湾近年来也有栽种。本研究探讨在宜兰和苗栗种植的忧遁草之生理功效及制成茶包冲泡茶汤之接受度。先比较溶剂萃取对忧遁草萃取物之功效;再比较宜兰与苗栗于不同採收季节的忧遁草及其乾燥条件对遁草甲醇萃取物生理活性之影响。忧遁草以热水、甲醇、乙醇萃取之萃取物,以甲醇萃取物的抗氧化能力及抑制醣苷酶、淀粉酶的能力最佳,甲醇萃取物的DPPH与ABTS清除能力为分别为6.77、36.82 mg TE/g extract (Trolox equivalent, TE);在浓度为5 μg/mL时抑制α-葡萄糖苷酶活性及α-淀粉酶活性的抑制率分别为63.55 %,11.36 %;Schaftoside的含量为2.099 mg/g DW (Dry weight)。种植在宜兰于冬季採收的忧遁草甲醇萃取物,其抗氧化能力及抑制醣苷酶及淀粉酶的能力比其他採收季节的高,其DPPH清除能力及ABTS清除能力分别为15.90、47.53 mg TE/g extract,Schaftoside含量为4.161 mg/g DW。抗氧化能力以种植在宜兰地区的比苗栗地区的佳。但苗栗地区的则以抑制醣苷酶及淀粉酶能力为最佳,其抑制α-葡萄糖苷酶活性的IC50为1.07 mg/mL,在浓度5 μg/mL时抑制α-淀粉酶活性能力为14.15 %。苗栗夏季採收的忧遁草,分别以冻乾、40 ℃、60 ℃、80 ℃热风乾燥、日晒乾燥,抗氧化能力以80 ℃乾燥的最佳,抑制醣苷酶及淀粉酶能力则以40 ℃乾燥的最佳;其抑制α-葡萄糖苷酶活性的IC50为0.91 mg/mL,在浓度5 μg/mL时抑制α-淀粉酶活性能力为16.82 %。忧遁草叶片以60 ℃乾燥制成茶包冲泡之茶汤,不论是在气味与口感方面,忧遁草茶汤整体喜好度皆能被消费者接受。


Clinacanthus nutans (C.nutans) is one of the traditional medicines in south east Asia (SEA). Recently, it has also been planted in Taiwan. This research discussed the physical effects of C.nutans and acceptance of consumers towards the teabags produced both in Yilan County and Miaoli County. First, the experiment was to compare the effects of solvent extraction on the extracts of C.nutans ; it was then to compare the effects of harvested in different seasons in Yilan and Miaoli and the drying conditions affected on the physiological activities of the methanol extract of C.nutans. C.nutans, along with hot water, methanol and ethanolic extract, the anti-oxidant ability and inhibition of glycosidase and amylase is the best in methanolic extract than that of the other. The scavengers of DPPH and ABTS are 6.77 mg TE/g extract (Trolox equivalent, TE) and 36.82 mg TE/g extract, respectively. The inhibitory ability of α-glucosidase and α-amylase at 5 μg/mL are 63.55% and 11.36%. Besides, the amount of the Schaftoside is 2.099 mg/g DW (Dry weight). The anti-oxidant ability and the inhibitory ability of glycosidase and amylase of methanolic extract is the highest in winter harvested than that of the other season. The scavengers of DPPH and ABTS are 15.90 mg TE/g extract and 47.53 mg TE/g extract, separately. The amount of the Schaftoside is 4.161 mg/g DW. On the other hand, the IC50 of inhibitory effect on α-glucosidase activity is 1.07 mg/mL. While the concentration is in 5 μg /mL, the inhibitory effect of α-amylase activity is 14.15%. Anti-oxidant ability of different drying temperature samples is stronger at 80℃ dried than that of the others, but the inhibitory ability of glycosidase and amylase is the sample dried at 40 ℃. The IC50 of α-glucosidase activity is 0.91 mg/mL. The concentration at 5 μg/mL to inhibit the activity of α-amylase is 16.82%. Therefore, the teabag of C.nutans, which is produced and dried at 60℃, not only creates delicate smell and taste, but also performs good overall preference, which can be generally accepted by consumers.
